This is also the first full-size game completely focused on the title character (Deadpool previously appeared as a supporting character in Marvel vs. Capcom 3: Fate of Two Worlds and Spider-Man: Shattered Divisions).

The script was written by Daniel Way, co-writer of comic series such as Wolverine: Origins, Deadpool and Starr the Slayer. The story begins with the kidnapping of the employees of High Moon Studios. The kidnapper turns out to be Deadpool, who demands that the developers create the best superhero game possible with himself as the main character.

In this game, players follow the adventures of the hero who takes the job to kill Chance White, a corrupt media mogul. However, his plans are interrupted by Mister Sinister, which costs Deadpool a lot of money. The mercenary wants revenge. Therefore, he agrees to cooperate with the X-Men and is sent to Genosha, an island of mutants, where Sinister headed to collect new DNA in order to create an army.
